Seromolecular Diagnosis of Avian Reovirus in Broiler Chickens: Analytical Interpretation of Field Cases

Abstract
Since the first report of avian reovirus (ARV) infection in Egypt in 1983, the virus has become endemic, with a significant negative impact on the poultry industry. This study was conducted to identify ARV in suspected flocks. For this purpose, eighteen chicken broiler flocks were investigated (October 2025 to May 2026). Initially, real-time reverse transcriptase polymerase chain reaction (rRT-PCR) was applied as the primary diagnostic method for viral detection. However, due to the low number of positive cases detected, enzyme-linked immunosorbent assay (ELISA) was subsequently used to further evaluate the infection status. Clinically affected flocks exhibited runting–stunting syndrome (RSS)/malabsorption syndrome, besides viral arthritis/tenosynovitis (VA) in ten flocks. rRT-PCR revealed the presence of ARV RNA in 6 flocks, but 10 flocks were positive for seroconversion using ELISA. Analysis of the results obtained from ARV-seromolecular diagnosis revealed several variable patterns; six flocks are positive for rRT-PCR, but ten flocks were positive for ELISA assay, including nine flocks with CV% ranging from 6.7% to 170.2%, indicating possible field infection. While four flocks evidenced positive for both rRT-PCR and ELISA. It could be concluded that the schematic diagnosis from day one of age with rRT-PCR on fecal swabs and/or tissue samples with paired serological monitoring to evaluate maternal immunity and confirm ARV field virus exposure as well as any other contributing agent.
