Comparative Effects of Topical Vitamin C and Zinc Oxide on the Healing of Skin Wounds in Rabbits: A Clinical and Histological Study

Abstract
Background: Wounds or injuries to the skin can be described as the whole or partial separation of the skin, and healing of such wounds is defined by a series of events that restore the skin to its normal state. Aims: This study evaluated the topical administration of vitamin C and zinc oxide on wound healing in full-thickness skin wounds, utilizing healthy male rabbits as the experimental model. Methodology: Eighteen healthy male rabbits were divided into three groups of six rabbits, each treated as follows: control (saline), vitamin C, or zinc oxide. Each rabbit in all three groups underwent surgical incision, creating a circular full-thickness wound (3 cm in diameter) on the dorsal thoracic region. All three groups received daily topical treatment from the study treatments for 21 consecutive days. The progress of wound healing was examined macroscopically and histologically on postoperative days 7, 14, and 21. Vitamin C and zinc oxide treatment improved the rates of wound contraction, epithelialization, and tissue regeneration at all time points compared with controls. Results: Vitamin C promoted earlier wound healing by reducing inflammation, presence of fibroplasias, and marked angiogenesis. Zinc oxide treatment showed a delayed response but had a higher number of favorable outcomes at the end of the study, including complete re-epithelialization, organized collagen, and complete formation of sebaceous glands and hair follicles. The histological examination of the zinc oxide-treated group exhibited the greatest degree of tissue remodeling at 21 days post-wounding. Conclusion: In conclusion, although both treatments had a beneficial effect on wound healing, zinc oxide-treated wounds demonstrated more complete restoration of skin architecture in the later stages of healing, suggesting it may be a more effective therapeutic agent for the treatment of open wounds.
