Abstract
Gastrointestinal nematode infections in dogs remain a significant concern due to their impact on animal health and their zoonotic potential. This study aimed to determine the prevalence and molecular phylogenetic characteristics of zoonotic gastrointestinal nematodes in captive dogs from an Indonesian captive animal facility. A cross-sectional study was conducted using fecal samples collected from 20 captive dogs immediately after defecation. Parasitological examination was performed using a flotation technique to identify nematode eggs, followed by fecal culture and Baermann methods to obtain infective third-stage larvae (L3). Morphological identification was complemented by PCR analysis of selected larval samples targeting the ITS-2 region, followed by DNA sequencing and phylogenetic analysis. Gastrointestinal nematode infections were detected in all examined dogs (100%), with Toxocara spp. and Ancylostoma spp. identified as the predominant parasites. The observed environmental conditions and management practices may have contributed to the persistence of parasite infections. Molecular analysis confirmed the presence of Ancylostoma ceylanicum and phylogenetic analysis showed that the obtained isolate clustered closely with reference sequences reported from several countries. In conclusion, the high prevalence of zoonotic gastrointestinal nematodes in captive dogs highlights the need for improved parasite control, regular monitoring, and enhanced sanitation in facilities with close human–animal interaction. The integration of morphological and molecular approaches strengthened species identification and confirmed the occurrence of A. ceylanicum in the studied captive dog population.

Key words: captive dogs, Ancylostoma ceylanicum, Toxocara spp., zoonoses, molecular phylogeny
