Molecular detection of certain enterotoxins of Clostridium perfringens isolated from canned fish retailed in Alexandria governorate

Abstract
Foodborne diseases (FBDs) remain a significant public health challenge in Egypt, particularly concerning resilient spore-forming pathogens like Clostridium perfringens. This study investigated the prevalence and molecular profile of C. perfringens in 120 canned fish samples (60 mackerel; 60 sardine) retailed in Alexandria governorate between September 2024 and February 2025. Biochemical identification revealed a uniform prevalence of 6.67% (4/60) for both mackerel and sardine samples. Molecular surveillance using PCR identified the species-specific cpa gene in eight isolates and the enterotoxin-encoding cpe gene in six isolates (four mackerel; two sardine). Sequence analysis of the cpe amplicons showed high homology (95.07%–97.65%) to the C. perfringens strain A1 CPE enterotoxin gene. These findings indicate a breach of the Egyptian Standard No. 287/2005, which mandates the total absence of anaerobic spore-formers in canned seafood. The high genetic identity among isolates suggests potential endemic contamination within local manufacturing or handling environments. This study underscores the necessity for rigorous molecular monitoring to ensure the safety of ready-to-eat canned products.
